Immune tolerance prevents the immune system from attacking the body's own tissues. Central tolerance removes self-reactive T cells in the thymus, while peripheral tolerance uses mechanisms such as T cell anergy, regulatory T cells (Tregs) and inhibitory receptors. Anergy occurs when a T cell receptor is stimulated without enough costimulation through CD28, leading to a state of unresponsiveness maintained by factors such as NFAT, EGR2, EGR3, DGK and the E3 ubiquitin ligases CBLB, ITCH and GRAIL (RNF128). Inhibitory receptors CTLA4 and PDCD1 (PD-1) and Treg cytokines IL-10 and TGF-beta also enforce tolerance, and failure of tolerance leads to autoimmunity.
