RAS proteins (KRAS, NRAS and HRAS) are small GTPases that act as molecular switches downstream of receptor tyrosine kinases. Growth factor receptors recruit GRB2 and the exchange factor SOS, which load RAS with GTP, while GTPase activating proteins such as NF1 switch RAS off. Active RAS stimulates the RAF-MEK-ERK (MAPK) pathway, PI3K-AKT, RALGDS and other effectors that drive cell proliferation, survival and differentiation. Activating RAS mutations occur in about 30% of human cancers, including pancreatic, colorectal and lung cancer.
