The tumor suppressor p53 (TP53), known as the guardian of the genome, is a transcription factor activated by DNA damage, oncogene activation, hypoxia and other stresses. Under normal conditions p53 is kept at low levels by the ubiquitin ligase MDM2. When stress kinases such as ATM, ATR and CHEK2 phosphorylate p53, it is stabilized and activates genes that stop the cell cycle (CDKN1A/p21, GADD45A), repair DNA, or trigger apoptosis (BAX, BBC3/PUMA, PMAIP1/NOXA, FAS) or senescence. TP53 is the most frequently mutated gene in human cancer.
