Toll-like receptors (TLRs) are pattern recognition receptors of the innate immune system that detect conserved microbial molecules. Cell surface TLRs (TLR1, TLR2, TLR4, TLR5, TLR6) recognize bacterial lipoproteins, lipopolysaccharide (LPS) and flagellin, while endosomal TLRs (TLR3, TLR7, TLR8, TLR9) recognize viral and bacterial nucleic acids. TLRs signal through the adaptors MYD88 and TRIF (TICAM1), activating IRAK kinases, TRAF6, NF-kappaB, MAPKs and IRF3/IRF7. This induces inflammatory cytokines such as TNF, IL1B and IL6 and type I interferons, and links innate to adaptive immunity.
