Schwann cells are the main glial cells of the peripheral nervous system. They wrap around axons to form the myelin sheath that allows fast nerve conduction, and they support axon survival and regeneration after injury. Schwann cell development and myelination are controlled by transcription factors such as SOX10, EGR2 (KROX20) and POU3F1 (OCT6), and by neuregulin 1 signaling through ERBB2/ERBB3 receptors. Myelinating Schwann cells express myelin proteins such as MPZ, PMP22 and MBP. Defects in Schwann cell genes cause peripheral neuropathies such as Charcot-Marie-Tooth disease.
