Rho family GTPases, including RHOA, RAC1 and CDC42, are molecular switches that control the actin cytoskeleton, cell polarity, migration, adhesion and cell division. They are activated by guanine nucleotide exchange factors (GEFs), inactivated by GTPase activating proteins (GAPs), and held inactive by guanine nucleotide dissociation inhibitors (GDIs). Active Rho GTPases signal through effectors such as ROCK, mDia, PAK and WASP to form stress fibers, lamellipodia and filopodia. Rho GTPase signaling is important in cancer invasion and metastasis, the immune response and neuronal development.
