The retinoblastoma protein (RB1) is a tumor suppressor that controls the transition from the G1 to the S phase of the cell cycle. In its hypophosphorylated form, RB1 binds E2F transcription factors and represses genes needed for DNA replication. Mitogenic signals induce cyclin D with CDK4/6 and cyclin E with CDK2, which phosphorylate RB1 and release E2F, allowing cells to enter S phase. CDK inhibitors such as CDKN2A (p16) and CDKN1A (p21) block this process. The RB pathway is disrupted in most human cancers, and CDK4/6 inhibitors are used in breast cancer therapy.
