Platelet derived growth factors (PDGF-A, -B, -C and -D) are growth factors for cells of mesenchymal origin, including fibroblasts, smooth muscle cells and pericytes. PDGFs bind the receptor tyrosine kinases PDGFRA and PDGFRB, causing receptor dimerization and autophosphorylation and activating the RAS-MAPK, PI3K-AKT, PLC-gamma and STAT pathways. PDGF signaling promotes cell proliferation, migration and survival and is important in embryonic development, wound healing and blood vessel maturation. Overactive PDGF signaling contributes to fibrosis, atherosclerosis and cancers such as glioma and gastrointestinal stromal tumors.
