Oxidative stress occurs when the production of reactive oxygen species (ROS), such as superoxide, hydrogen peroxide and hydroxyl radicals, exceeds the cell's antioxidant capacity. ROS are made by mitochondria, NADPH oxidases (NOX) and other enzymes, and can damage DNA, proteins and lipids. Cells defend themselves with antioxidant enzymes including superoxide dismutases (SOD1, SOD2), catalase, glutathione peroxidases (GPX), peroxiredoxins and thioredoxins. Many of these genes are induced by the NRF2 (NFE2L2)-KEAP1 pathway. Oxidative stress is involved in aging, cardiovascular disease, neurodegeneration, diabetes and cancer.
