The Notch signaling pathway is a cell-cell communication system that controls cell fate decisions, proliferation and differentiation during development and in adult stem cells. Notch receptors (NOTCH1-4) on one cell bind Delta-like (DLL1, DLL3, DLL4) or Jagged (JAG1, JAG2) ligands on a neighboring cell. This triggers cleavage of the receptor by ADAM proteases and gamma-secretase, releasing the Notch intracellular domain (NICD), which enters the nucleus and works with RBPJ and MAML to activate target genes such as HES1, HEY1 and NRARP. Abnormal Notch signaling is linked to leukemia, solid tumors and developmental disorders.
