Myeloid derived suppressor cells (MDSCs) are immature myeloid cells that expand in cancer, chronic inflammation and infection and strongly suppress T cell and NK cell responses. MDSCs are divided into monocytic (M-MDSC) and polymorphonuclear (PMN-MDSC) subsets. They are expanded and activated by factors such as GM-CSF, G-CSF, IL-6, VEGF and S100A8/A9 acting through STAT3 and NF-kappaB. MDSCs suppress immunity through arginase 1 (ARG1), inducible nitric oxide synthase (NOS2), reactive oxygen species, IL-10 and TGF-beta, and are an important barrier to cancer immunotherapy.
