Type I interferons (IFN-alpha and IFN-beta) are produced in response to viral infection when pattern recognition receptors such as TLRs, RIG-I (DDX58), MDA5 (IFIH1) and cGAS-STING detect viral nucleic acids. Type I IFNs bind the IFNAR1/IFNAR2 receptor, activating JAK1 and TYK2, which phosphorylate STAT1 and STAT2. Together with IRF9 they form the ISGF3 complex, which induces hundreds of interferon stimulated genes (ISGs) such as MX1, OAS1, ISG15 and IFIT genes that block virus replication. Type I interferon signaling also shapes adaptive immunity and is overactive in autoimmune diseases such as lupus.
