Hypoxia signaling allows cells to adapt to low oxygen levels. Under normal oxygen, the hypoxia inducible factor alpha subunits (HIF1A, EPAS1/HIF2A) are hydroxylated by prolyl hydroxylases (EGLN/PHD), recognized by the VHL ubiquitin ligase and degraded. Under hypoxia, HIF-alpha is stabilized, pairs with ARNT (HIF1B) and activates genes that increase oxygen delivery and anaerobic metabolism, such as VEGFA, EPO, SLC2A1 (GLUT1), glycolytic enzymes and CA9. Hypoxia signaling is important in development, ischemia, inflammation and tumor growth and angiogenesis.
