The Hippo signaling pathway controls organ size, tissue regeneration and stem cell function by regulating cell proliferation and apoptosis. The core of the pathway is a kinase cascade in which MST1/2 (STK4/STK3) with SAV1 phosphorylate LATS1/2 with MOB1. LATS1/2 then phosphorylate the transcriptional coactivators YAP1 and TAZ (WWTR1), keeping them in the cytoplasm. When Hippo signaling is off, YAP/TAZ enter the nucleus and bind TEAD transcription factors to induce growth genes such as CTGF and CYR61. The pathway responds to cell density, cell polarity and mechanical cues, and its dysregulation contributes to cancer.
