Fibroblast growth factors (FGFs) signal through four receptor tyrosine kinases, FGFR1 to FGFR4, with heparan sulfate proteoglycans and, for endocrine FGFs, Klotho coreceptors. Activated FGFRs phosphorylate the adaptor FRS2 and PLC-gamma, leading to activation of the RAS-MAPK, PI3K-AKT and STAT pathways. FGFR signaling regulates embryonic development, tissue repair, angiogenesis and phosphate and bile acid metabolism. FGFR amplifications, mutations and fusions drive several cancers, and FGFR mutations cause skeletal disorders such as achondroplasia.
