BRCA1 is a tumor suppressor that maintains genome integrity through homologous recombination DNA repair, cell cycle checkpoint control and transcriptional regulation. BRCA1 works with BRCA2, PALB2, RAD51, ATM, ATR and CHEK2 to detect and repair DNA double-strand breaks, and inherited mutations in BRCA1 or BRCA2 greatly increase the risk of breast and ovarian cancer. Breast cancer signaling also involves estrogen and progesterone receptors, HER2 (ERBB2), PI3K-AKT and MAPK pathways, which drive proliferation and survival of breast tumor cells.
