Bone morphogenetic proteins (BMPs) are members of the TGF-beta superfamily that regulate embryonic development, bone and cartilage formation, and tissue homeostasis. BMP ligands bind type I (BMPR1A, BMPR1B, ACVR1) and type II (BMPR2) serine/threonine kinase receptors, leading to phosphorylation of SMAD1, SMAD5 and SMAD8. These receptor-regulated SMADs partner with SMAD4 and move to the nucleus to control target genes such as the ID genes. BMP signaling is fine-tuned by extracellular antagonists such as noggin, chordin and gremlin and by inhibitory SMAD6 and SMAD7, and its dysregulation is linked to skeletal disorders, pulmonary arterial hypertension, fibrosis and cancer.
